Brief Statement of research work

My research is a decade-long translational program converting molecular discoveries into public, economic, and environmental benefits, advancing UN Sustainable Development Goals. I investigated molecular pathophysiology of metabolic disorders, effects of environmental toxicants on metabolism, and pragmatic natural-product therapeutics with formulation science. The program links mechanistic discovery, therapeutic innovation, policy engagement, and capacity building to address Pakistan diabetes burden. My portfolio includes seven competitively funded multidisciplinary projects, creating a pipeline from lab discovery to application. A gender-specific biomarker study produced Pakistan first evidence of sex differences in diabetic inflammation, enabling gender-responsive screening. BPA research linked endocrine disruption to T2DM, evaluated plant candidates, and informed regulatory reform (SDG-12). Complementary work validated local bioactives and antidiabetic efficacy of Allium cepa and Zingiber officinale oils, supporting nutraceutical and cottage-industry production (SDG-3, SDG-8). Nicotine studies revealed extra-pancreatic mechanisms, strengthening tobacco-control advocacy. Prediabetes modeling produced predictive screening tools for Pakistan and Saudi Arabia. Heavy-metal susceptibility data informed water-safety and waste-management policies (SDG-6, SDG-11). We developed polymer delivery platforms to improve oral stability and bioavailability and filed a patent for an Alcalase-encapsulated chitosan probiotic matrix. With broad scholarly output and institutional leadership, my work delivers biomarkers, predictive tools, interventions, trained researchers, and industry-ready prototypes with national impact.

Beyond pancreas: Investigation of novel targets and treatment for nicotine-induced diabetes mellitus

This project found that chronic nicotine exposure disrupts glucose homeostasis by inducing insulin resistance, ??-cell dysfunction, oxidative stress, inflammation, and mitochondrial impairment. Untargeted metabolomics revealed consistent amino-acid and lipid pathway perturbations linked to these defects. Preclinical interventions with resveratrol normalized key metabolic and metabolomic markers, reduced oxidative/inflammatory burden, protected organs, and improved glycaemia and insulin sensitivity. Outcomes identify non-pancreatic targets (redox, inflammatory and mitochondrial pathways), support nutraceutical mitigation strategies, and justify translational evaluation alongside policies to reduce nicotine exposure.

Heavy Metal Induced Disease Susceptibility

This project investigated mechanistic links between heavy-metal exposure and disease susceptibility. Integrated biochemical, toxicological, and molecular analyses demonstrated that arsenic and related metals induce oxidative stress, mitochondrial dysfunction, and pro-inflammatory signaling, thereby impairing glucose and lipid metabolism and increasing cardiometabolic risk. Experimental models showed disruption of insulin signaling pathways, altered antioxidant defenses, and tissue-specific metabolic derangements. Biomarker profiling identified redox and inflammatory indices as early indicators of toxicity. Importantly, ed plant-derived bioactives attenuated metal-induced metabolic and oxidative abnormalities. These findings provide mechanistic evidence for heavy-metal???associated disease vulnerability and support preventive strategies, exposure control policies, and development of protective nutraceutical interventions.

Evaluation of risk factors associated with the development of prediabetes

The DSR2022-RG-0144???supported studies elucidated metabolic and genetic risk factors linking prediabetes with cardiometabolic complications. Clinical analyses confirmed that diabetes and hypertension significantly increase the risk of adverse outcomes, including pre-eclampsia and cardiovascular events. Molecular investigations demonstrated that the MTHFR C677T polymorphism is associated with altered homocysteine metabolism and heightened myocardial infarction susceptibility in diabetic patients. Additionally, regulatory variants of KLF14 were significantly associated with dyslipidemia, insulin resistance, and cardiodiabetic phenotypes. Collectively, these findings integrate clinical and genetic evidence to refine risk stratification, enabling early identification of high-risk individuals and supporting precision-based preventive strategies.

Investigations of therapeutic potentials of Allium cepa and Zingiber officinale essential oils on various pathways involved in diabetes mellitus

This project showed that essential oils of Allium cepa and Zingiber officinale provide complementary antidiabetic actions. In vitro and animal studies demonstrated ??-amylase and ??-glucosidase inhibition, antioxidant and anti-inflammatory effects, enhancement of insulin signaling and GLUT4 translocation, and improved mitochondrial function, cumulatively reducing hyperglycaemia and insulin resistance. Phytochemical profiling identified sulfur compounds, flavonoids, and gingerols as active constituents. Preclinical intervention reduced oxidative markers and normalized key metabolic biomarkers. Outcomes support development of onion and ginger essential-oil???based nutraceuticals and justify progression to clinical evaluation and formulation optimization for diabetes adjunct therapy.

Biochemical investigation of association of endocrine disrupting chemicals with insulin-stimulating and carbohydrate metabolizing hormones in diabetes mellitus and their treatment strategies using plant-based bioactive compounds

This project demonstrated that EDCs disrupt insulin-stimulating and carbohydrate-metabolizing hormones, contributing to impaired glucose homeostasis. Combined human, animal, and biochemical studies linked arsenic and BPA exposure to altered insulin, glucagon, and incretin signaling, elevated insulin resistance, and modulation of ??-amylase/??-glucosidase activity. Metabolomics and mechanistic assays implicate oxidative stress, inflammation, and mitochondrial dysfunction as mediators. In vitro and in vivo screening identified plant-derived bioactives (resveratrol, taxifolin) that inhibit carbohydrate-metabolizing enzymes, restore hormonal and metabolic markers, reduce oxidative/inflammatory load, and improve glycemic outcomes - supporting nutraceutical strategies and exposure-reduction policies. Findings warrant clinical translation and policy action nationwide in Pakistan.

Bisphenol A - associated type 2 diabetes mellitus: Pathogenesis and prospects for treatment

This project integrated epidemiology, controlled animal studies, metabolomics, and interventional experiments to elucidate how chronic BPA exposure promotes T2DM and to evaluate natural-product mitigants. Environmentally relevant BPA doses disrupted glucose homeostasis by inducing insulin resistance, impairing ??-cell function, and altering adipokine and lipid metabolism. Metabolomics revealed perturbations in amino acid, fatty acid, and energy pathways consistent with mitochondrial dysfunction and oxidative stress. Mechanistic analyses implicated inflammatory signaling and redox imbalance. Resveratrol and related phytochemicals reversed metabolic abnormalities and improved insulin sensitivity, supporting translational mitigation strategies and evidence-informed policy to reduce BPA exposure.

Gender-specific association between serum inflammatory biomarkers and insulin resistance in diabetic patients

This research funded by HEC examined serum IL-6, TNF-?? and CRP in Pakistani type 2 diabetic patients to determine sex-specific links with insulin resistance. In a cross-sectional cohort, higher IL-6, TNF-?? and CRP correlated with HOMA-IR, with associations markedly stronger in women and partially mediated by greater BMI and central adiposity. Multivariate analyses and mechanistic interpretation indicate sexual dimorphism in inflammatory contributions to insulin resistance. These findings support incorporating inflammatory biomarkers into diabetes risk stratification and endorse sex-tailored prevention and therapeutic strategies targeting inflammation and adiposity to reduce diabetes progression nationwide in Pakistani clinical settings.
